A consortium of retinal specialists has issued a caution regarding the potential side effects of Syfovre, a newly approved ocular medication, suggesting that these effects may be associated with an elevated risk of inflammation that could result in blindness in certain instances.
Syfovre (Pegcetacoplan injection) was launched by Apellis Pharmaceuticals in February 2023, following its approval by the FDA for the treatment of geographic atrophy, a prevalent cause of vision impairment. However, as the medication gains traction among patients, there have been reports of significant adverse effects linked to Syfovre, which may not have been sufficiently communicated by the manufacturer.
Earlier this month, the American Society of Retinal Specialists (ASRS) dispatched a letter to fellow healthcare professionals, alerting them that Syfovre has been associated with at least six instances of occlusive retinal vasculitis, a severe condition that obstructs blood flow to the retina and can result in eye inflammation or total vision loss.
A report from BioPharma Dive indicates that the ASRS letter cautions that eye inflammation side effects associated with Syfovre may manifest within one to two weeks following a patient’s initial injection of the medication, which is generally administered every one to two months.
The ASRS observed that the eye inflammation issues related to Syfovre did not seem to be associated with any particular batch of the drug, and the underlying cause of these side effects remains uncertain. Nevertheless, the group of retinal specialists has advised healthcare providers to remain vigilant, ensure thorough follow-up for patients who have recently received a Syfovre injection, and encouraged both doctors and patients to report any adverse events to the FDA. Officials from Apellis have stated that they are currently investigating these incidents.
In July 2023, the Research and Safety in Therapeutics Committee of the American Society of Retinal Specialists (ASRS) disseminated a communication to its members, highlighting numerous reports of retinal vasculitis and retinal vascular occlusion in individuals who had undergone the Syfovre injection.
As of December 1, 2023, Apellis Pharmaceuticals, Inc. confirmed the occurrence of 12 cases of retinal vasculitis, along with two additional suspected instances. Several patients who received Syfovre injections experienced intra-ocular inflammation (IOI) of varying severity, as well as retinal vasculitis and occlusive retinal vasculitis following a single administration of Syfovre.
